Uluslararası Dergilerde Hakemlik Rapor Örneği Editor and reviewer comments: --Reviewer 1 In the present study, the role of XXX receptors (XX) in the neuroprotective effect of estrogen were investigated after traumatic brain injury (TBI). 105 ovariectomized animals were divided into 5 groups including Control (TBI), Estrogen, XX 1 -, XX 2 -, and XX 3 inhibitors. Main problem of this study is the XXX receptor inhibitors were not used alone. What we know about inhibitors that almost all kinds of inhibitors effect signaling pathways, which may modulate or activate survival kinases such as Akt signaling components. In this context, although XXX was not used in this study, endogenous XXX synthesis should also be considered. In addition, I do not understand exact hypothesis of this study. The reasons of the combination of XXX receptor antagonists and estrogen are not convincing enough, especially from this study design without XXX. Especially inhibitors alone groups were not included to this study, causing further questions. From this study design and methods we will not be sure estrogen bindings to XX receptors. Binding analyzes should be done. In addition, the effect of estrogen is more complicated, its classical receptors are in the cytoplasm but it has novel membrane receptors. And we will be not sure whether these XX receptors inhibitors have un-specific binding effect on these membrane receptors. In addition endogenous XXX synthesis and the effect of estrogen on XXX synthesis should be also concidered. Luzindole is not specific to XX 1, it is also a XX 2 antagonist, There is no text about Western blot in the methods. Treatment routes of XX receptor inhibitors are not clear. Animal groups should be defined in materials and methods more preciselly. It is difficult to understand the first 2 groups from 4 subtitles. They should be 5 subtitles. Manuscript should be edited, e. g. hoursafter; mg/kg. Evans (Page 9). Taken together this study is not well designed. The mechanism of estrogen-XXX interactions are complicated and needs complicated study design.
