Pollen preparation method • • • Boil the sample in 10% KOH Sieve Wash the material Dehydrate with acetic acid Acetolyse the material by heating the sample in the acetolysis mixture to 100 °C. Acetolysis dissolves most of the tissue and organic debris, and removes the proteins, lipids, and carbohydrates from the surface of the pollen grains. This makes the pollen grains easier to stain, photograph, and identify. Cool the sample tubes, wash with distilled water alcohol. The separation of organic material from sand clay Put the residue tube for a night in the oven at 40 °C. Prepare microscope slides.
